What's a good pumping schedule for building up a supply of expressed milk?

Are you currently breastfeeding as well?

When I was exclusively pumping I pumped every 3 hours with one 6 hour break in a day (usually at night!). I was up to 2 oz a side in no time....

Make sure to pump first thing in the morning, since nightime/sleeptime is when prolactin levels are at their highest. If baby nurses on one side only, pump the other. The key is, an empty breast at the end of a feeding/pumping session signals the production of more milk. The breast is never truly "empty", and remember, it is a supply and demand operation. Increase the demand, the body will accommodate. It is going to take some commitment from you, even when tired, to pump after you nurse. It's worth it though!
